yes they showed us they got the tattoo on the ep but they must have go it before the ep I would have 사랑 to see that part were they went to a tattoo 샵 and got it done that would have been awesome to see.
We don't see them getting the 문신 또는 know exactly when they got them. However, it must have been between 심장 in S2 (you can see Sam doesn't have it yet) and Jus In Bello in S3, when they first show the tattoos.
They didn't get the tattoo in an episode of Supernatural, but when they were much younger. Except, Sam got it of 의해 the Help of Castiel, so that Crowley would be able to posses him to remove Gadreel from Sammy (9.10). So, now, I do not know if Sammy got it back 또는 not

We know they didn't have them pre-series because Sam gets possessed in season 2. We also saw shirtless scenes of both brothers in the first two seasons where they were tattoo-less.
